The locations are one difference, while not far apart (Singapore city is quite compact): depending on what you enjoy, the Singapore Hilton is on Orchard Road and would be very convenient to shopping up and down Orchard, if that is how you wanna spend your time.
The Conrad is more downtown and closer to the cricket club fields and government buildings and closer to Boat Quay and Clarke Quay, which are lively, but focused primarily on restaurants and pubs (especially Boat Quay, with 70+ various ethnic restaurants along the water all in a row) and it's also closer to Chinatown and the new vertical center-city malls at SunTec Center. Little India (great veg. restaurants) in the Bugis Junction area is closer to the Conrad, too.
As for the hotels themselves, with your stay limited to two nights, I'd opt for the Hilton on Orchard, because cabs are inexpensive once you decide what you'll do with your time and most hotels in the city are comfortable by any standard.

Hilton Singapore is located very close to the famous Singapore shopping belt - Orchard Road. If you are looking to spend sometime in Singapore to do some serious shopping, Hilton would be the choice. The Conrad Singapore on the other hand is located in the heart of Singapore's busiest business district and Marina Bay Suntec Singapore. The hotel is in close proximity to Singapore's central business district and 20min drive from Singapore Changi Airport. Conrad would be ideal for visitors on a business trip in Singapore.
