My husband and I will be headed to London (8 nights), Killarney (3 nights) and Dublin (3 nights) in just under two months.
We're putting the final touches on the details of our trip - we already have hotels/B&Bs and most of the transportation worked out. Last week, we wrote our letter to the Tower of London requesting tickets to see the Ceremony of the Keys.
Are there any other sites (museums, tours, shows, transportation) we should try to get tickets to in advance as opposed to when we arrive in October?
We ended up purchasing our London Eye tickets in advance on-line, show tickets, and I think we even purchased tickets to one of those hop-on, hop-off buses ahead but it was the show and the eye tickets I was glad we got in advance.
Depending on where you want to have dinners, it may be a good idea to book tables in advance. It's often difficult to end up at certain restaurants if one doesn't do that at least a few weeks ahead.

If you're thinking of visiting the Guinness museum/exhibition in Dublin (known as the Storehouse), advance tickets are a little cheaper. Having said that, the Jamieson whisky museum is probably better, although the view from Guinness's Gravity bar is impressive.
An idea for you. The Abbey Theatre is worth going to on a cold evening. Their site shows Lay Me Down Softly is on about the time you will be there. I don't think you will need to book in advance, although the performance we saw was a preview, so normal performances may be more booked.
