We will be staying in Tripolli, Greece for one week and then to Positano for a week. Could someone tell me the best way to travel across the waters. Plane, train, boat? We don't want to waste too much time.

Probably your best option is a flight. Check out this no frills airline, connecting Athens and Naples: Alpi Eagles.
