We will be travelling from Siena to Rome via Sena bus. The trip is about 3 hours. Does anyone know whether the bus makes a long stop
(toilet break) or do the buses have toilets on board?

I did a search on this forum with the words "bus" and "toilet" and found an old thread and a reply from Cristina that there is no toilet on the bus.

However --- are you sure this is a 3-hour trip? Seems VERY long for that distance. I've checked it out recently, and think it's about 1 1/2 hours.

I'd check it myself on the SENA website, but I can't seem to get it to work.

I've ridden the blue intercity busses from Assisi to Siena, from Siena to Florence, from Sorrento to Salerno, and from Rome to Ariccia but have never seen a toilet on board.

The Assisi to Siena trip had a stop in Perugia, and the Sorrento to Salerno stopped in Amalfi.

Yes, it is a three-hour trip. I haven't gone on that route, but in my experience buses do make pit stops on trips of that length.

I've done the bus trip a few times, the first time from Siena to Rome. It was a really nice bus; it must have been brand new, with a toilet at the back entrance, air conditioning -- really comfortable for what is a fairly long trip.

But last fall, when I took my elderly mom in the other direction out of Rome, it wasn't great. No toilet, no pitstop, no aircon, really grubby seats with broken arm rests.

So in my limited experience, it seems to be a bit of a crap shoot in terms of what you get! But it may be, that if you actually ask the driver to make a stop midway, he might be obliging. And there is some lovely scenery on the route, which helps the three hours pass fairly quickly.

I did the Sena bus trip Siena to Rome recently. I don't think there was a toilet on board. However there was a pitstop for about 10 minutes at a half way point.The bus was clean.
did you know the bus terminates at Rome Tiburtina station. Then you need to catch the Metro B line to Rome Termini if that's where you want to go.

Yes I know the bus goes to Tiburtina. This suits us as we then go by metro to Cavour.
