The route is clearly marked. The rental agencies are on the right as you come off the causeway from Mestre - just before the Piazzalle Roma. You park the car in the garage and then go to the rental office. As for gas - my recollection is that the area in Mestre just before the causeway is pretty industrial.So you should find gas stations. I believe there is also gas available on the Venice side. Perhaps someone else who has done this more recently may have specific directions for gas. You might want to indicate where you are coming from.

We usually fill up on gas at the last service station on the Autostrada before the exit to Venice. We have returned cars 4 times in Venice doing this w/o any problems. The signs from the Autostrada exit to Venice are large and frequent. You should not have a problem. Sometimes the car return area is extremely crowded and parking the car can be a challenge. Just do the best you can. Be careful crossing the street towards the vaporetto stop. The cars do whiz by and you will be dragging your luggage.

I jsut returned a rental in Venice on April 21st.
There is a gas station on your left as you come off the causeway. No problem getting in or out of it and prices were no higher than on the mainland. This was for diesel fuel.
There is no official parking place for rental returns; just park somewhere in front of all the rental offices on the main road.
